Mechanisms of CTL Resistance in Persistent HIV Reservoirs

Antiretroviral therapies (ART) durably suppress HIV replication, but leave a reservoir of infected cells that re-seed viremia if ART is interrupted. The ability of these infected cells to avoid immune clearance has been attributed to viral latency, where integrated proviruses remain antigenically silent. However, converging lines of recent evidence point to a second mechanism of resistance comprising cells that express HIV but are intrinsically resistant to elimination by cytotoxic T cells. In this seminar I will review this evidence and present novel insights into mechanisms of resistance – with an eye towards novel therapeutic strategies to cure HIV.
